ABSTRACT

Objective:To explore the expression of long non-coding RNA (lncRNA) CDK5RAP3 in gastric cancer tissue and its regulatory effect on gastric cancer cell proliferation and invasion.Methods:The expression differences of CDK5RAP3 in gastric cancer tissues and adjacent tissues were analyzed by TCGA database. By transfecting the pcDNA3.1-CDK5RAP3 plasmid into Hs-746T cells, a gastric cancer cell line overexpressing CDK5RAP3 (CDK5RAP3 group) was constructed, and the pcDNA3.1 plasmid was transfected into Hs-746T cells as a control group. The changes of CDK5RAP3 expression in the two groups of cells were detected by real-time quantitative PCR (qRT-PCR). The effects of overexpression of CDK5RAP3 on the proliferation and invasion of Hs-746T cells were detected by CCK-8 assay and Transwell assay, respectively. The binding sites of CDK5RAP3 and miR-223-3p were predicted by the starBase v2.0 database. The direct binding of CDK5RAP3 and miR-223-3p was verified by dual-luciferase reporter gene experiment. The expression levels of miR-223-3p in Hs-746T cells in each group were detected by qRT-PCR. Western blot was used to detect the expression levels of proliferation proteins and invasion proteins in Hs-746T cells in each group. The experimental data were analyzed by SPSS 17.0 software, and the measurement data conforming to the normal distribution were expressed as Mean±SD. The t-test was used to compare between two groups, and the one-way analysis of variance was used to compare the means of multiple groups. Results:Compared with adjacent tissues, the expression level of CDK5RAP3 in gastric cancer tissues was significantly lower ( P<0.01). The expressions of CDK5RAP3 in Hs-746T cells in the control group and CDK5RAP3 group were (1.08±0.77) and (10.63±2.14), respectively, and the difference was statistically significant ( P<0.01). Up-regulation of CDK5RAP3 significantly decreased the proliferation activity of Hs-746T cells ( P<0.05). The number of invasive cells in the control group and CDK5RAP3 group were (137.80±28.72) and (57.76±24.95), respectively, and the difference was statistically significant ( P<0.01). CDK5RAP3 could directly bind miR-223-3p ( P<0.01). The expression of miR-223-3p in Hs-746T cells in control group and CDK5RAP3 group were (6.22±1.20) and (1.01±0.98), respectively, and the difference was statistically significant ( P<0.01). Compared with the control group, up-regulation of CDK5RAP3 significantly reduced the expression levels of proliferation and invasive proteins. Conclusion:The expression of CDK5RAP3 is low in gastric cancer tissue, and CDK5RAP3 inhibits the proliferation and invasion of gastric cancer Hs-746T cells by targeting miR-223-3p.

ABSTRACT

Object the current radiological health situation of different ranks medical institutions of interventional radiology in a city of Pearl Delta. Methods all the medical institutions in the city, which had carried out interventional radiology before December 31, 2019 as the research objects by survey. By using the method of field epidemiological investigation, the basic situation of general messages, the status of protective facilities, environmental radiation does, personal protective equipment, occupational health monitoring were investigated. Results There were 12 interventional radiology equipment and 180 employees in 7 medical institutions in the city, before December 31, 2019. There was no statistical significance in the composition of employees of different ranks medical institutions (P > 0.05). There was no statistical significance in the composition of protective facilities for interventional treatment in different ranks hospitals (P > 0.05). The environmental radiation dose in interventional radiology devices room of grade A class 3 hospital was higher than that of grade A class 2 hospital (P < 0.05). There was no statistical significance in the composition of individual protective equipment for interventional therapy in different ranks medical institutions (P > 0.05). The proportion of occupational health examination before work and during work in grade A class 2 hospital was higher than A class 3 hospital (P < 0.01). Conclusion The current situation of radiological health of different ranks medical institutions of interventional radiology in a city of Pearl Delta was good momentum. It may be helpful to improve the radiological health protection level of interventional therapy in this city by strengthening the radiological protection management of applying for interventional therapy, actively carrying out the evaluation of radiological protection against occupational disease hazards in construction projects, and urging the employee to carry out the occupational health monitoring inspection as required.

ABSTRACT

DNA is the hereditary material in humans and almost all other organisms. It is essential for maintaining accurate transmission of genetic information. In the life cycle, DNA replication, cell division, or genome damage, including that caused by endogenous and exogenous agents, may cause DNA aberrations. Of all forms of DNA damage, DNA double-strand breaks (DSBs) are the most serious. If the repair function is defective, DNA damage may cause gene mutation, genome instability, and cell chromosome loss, which in turn can even lead to tumorigenesis. DNA damage can be repaired through multiple mechanisms. Homologous recombination (HR) and non-homologous end joining (NHEJ) are the two main repair mechanisms for DNA DSBs. Increasing amounts of evidence reveal that protein modifications play an essential role in DNA damage repair. Protein deubiquitination is a vital post-translational modification which removes ubiquitin molecules or polyubiquitinated chains from substrates in order to reverse the ubiquitination reaction. This review discusses the role of deubiquitinating enzymes (DUBs) in repairing DNA DSBs. Exploring the molecular mechanisms of DUB regulation in DSB repair will provide new insights to combat human diseases and develop novel therapeutic approaches.

ABSTRACT

Objective To evaluate the efficacy of reinforcing kidney and removing stasis combined with thalidomide in the treatment of multiple myeloma.Methods 120 patients with multiple myeloma from January 2011 to January 2015 in department of traditional Chinese medicine of the third hospital affiliated to Liaoning medical university were divided into two groups.The control group received VAD program ( vincristine, doxorubicin and dexamethasone), and the observation group received reinforcing kidney and removing stasis combined with thalidomide on the basis of control group.All patients received treatment for four weeks of one course.Results After treatment, the total efficiency of observation group was higher than that of control group(75% vs.60%, P<0.05).Compared with control group, the M protein,plasmocyte count and osteoclasts count of observation group were lower, the osteoblast count of observation group was higher(P<0.05).The improvement rate of quality of life (KPS score) in observation group was higher than that in control group (70.00% vs.51.67%,P<0.05).The changes of renal function (serum creatinine, urea nitrogen) and adverse reaction in observation group were less than those in control group (P<0.05).Conclusion The reinforcing kidney and removing stasis combined with thalidomide has the significant effect in the treatment of multiple myeloma with fewer adverse reactions and higher quality of life after treatment.

ABSTRACT

Background: Vascular endothelial growth factor [VEGF]-C, as a lymphangiogenic factor, plays important roles in the progression of several malignancies. However, its clinical prognostic value in breast cancer still remains controversial. We performed a meta-analysis of available studies to assess the association between VEGF-C expression and the outcomes of breast cancer patients


Results: We searched eligible studies in three English databases [MEDLINE, EMBASE, and Web of Science] and two Chinese databases [Wanfang and Chinese National Knowledge Infrastructure databases]. Key words used in the research included 'VEGF-C", "breast cancer", "immunohistochemistry", "breast neoplasma[s]", "breast carcinoma", "metastasis", and "prognosis". Fourteen studies with a total of 1, 573 breast cancer cases were finally included into the meta-analysis. The pooled odds ratios [ORs] with the corresponding 95% confidence interval [95% CIs] for lymph node metastasis, overall survival, and disease-free survival were calculated by using fixed-effects or random-effects models. Heterogeneity and publication bias were also assessed


Results: Meta-analysis of random-effects model showed VEGF-C expression was associated with lymph node metastasis in patients with breast cancer [random-effects, OR = 2.14; 95 % CI 1.21-3.77, P = 0.009]. VEGF-C expression was associated with poorer overall survival [fixed-effects, OR = 2.46, 95% CI: 1.46-4.14, P < 0.001] and disease-free survival [fixed-effects, OR = 2.10, 95% CI: 1.32-3.35, P = 0.002] in patients with breast cancer


Conclusion: VEGF-C expression is positively associated with lymph node metastasis in breast cancer, and VEGF-C detection in breast cancer might be an effective and feasible means to predict outcome

ABSTRACT

ObjectiveThe aim of this study is to evaluate diagnostic and prognostic value of matrix metalloproteinase-7 (MMP-7) in patients with bladder cancer.MethodsA total of 77 patients with bladder cancer and 65 healthy controls were recruited in this study.The levels of serum MMP-7 were measured by ELISA methods.ResultsThe level of serum MMP-7 in bladder cancer group was significantly higher than those in the healthy control [ [ 3.1 (0 - 32.1 ) ng/ml vs 1.0 ( 0.2 - 2.6) ng/ml,x2 =9.29,P <0.01 ].The level of serum MMP-7 was correlated to lymph node metastasis ( x2=10.49,P < 0.01 ) and tumor grade ( x2=4.55,P =0.033 ).Receiver operating characteristic ( ROC ) curves of serum MMP-7level was 0.810.Serum MMP-7 was used to diagnose bladder cancer with the sensitivity of 83.6% and specificity of 77.4%.The survival rate in patients with a high level of MMP-7 was lower than that with a low level of MMP-7 (x2=3.88,P =0.0488).ConclusionsMMP-7 may be closely associated with the progression of bladder cancer.Serum MMP-7 may be a useful index for diagnosis and prognosis for patients with bladder cancer.

ABSTRACT

ObjectiveTo observe the potential effects of microcystin-LR (MC-LR) to open field behaviors and cognitive functions of rats after low-dose and 60 days exposure.Methods20 SD male rats (4 weeks old) were grouped into control ( double distilled water),0.2,1.0 and 5.0 μg/kg groups by random number table.Rats were exposed to microcystin-LR intragastrically every two days for 60 d.Open fields and Morris water maze were employed to observe the spontaneous behavior and spatial learning and memory capacity of rats.ResultsFor each group,the duration in the central area of open field were ( 8.86 ± 3.38 ) s,( 3.09 ± 1.22 ) s,( 1.68 ± 0.24) s,(1.22 ±0.93) s,frequencies in central area were (4.80 ± 1.16) times,(4.40 ± 1.91 )times,( 1.40 ±0.23)times,(3.80 ± 1.02 ) times,movement duration were ( 137.30 ± 5.25 ) s,( 122.97 ± 9.50) s,( 108.86 ± 6.38 ) s,( 129.35 ± 9.40) s,the escape latency were ( 23.17 ± 4.28 ) s,(77.00 ± 8.31 ) s,(59.00 ± 11.89) s,( 63.80 ±16.09) s,respectively.Compared to control,the duration in the central area of open field decreased and escape latency increased significantly (P < 0.05 ) in all the MC-LR treated groups; and frequencies in central area and movement duration of 1.0 μg/kg treated rats decreased significantly.No significant differences were observed between expsoured group and control group in spacial probe tests(P>0.05).ConclusionMC-LR exerts effects on the spontaneous behavior and spatial cognitive ability of rats.The further study on neurobehavior effects of MC-LR is necessary.

ABSTRACT

Objective The aim of this study is to assess the value of serum vascular endothelial growth factor-C (VEGF-C) for predicting recurrence and prognosis in patients with esophageal cancer.Methods A total of 65 patients with esophageal cancer,40 healthy controls were recruited in this study.Serum levels of VEGF-C were measured by ELISA methods.Results Serum level of VEGF-C in esophageal cancer group was higher significantly than those in controls [7.2 (1.2 ~ 35.5) μ g/L vs.4.2 (0.2 ~ 12.6)μ g/L,P < 0.01].Receiver operating characteristic (ROC) curve of serum VEGF-C for predicting recurrence was 0.781 (P< 0.01); sensitivity and specificity were 73.1% and 61.5%,respectively.The survival rate in patients with high VEGF-C level was lower than those with low VEGF-C level (P < 0.01).Conclusions VEGF-C may be closely associated with the development and metastasis of esophageal cancer.Serum VEGF-C is a useful marker for predicting recurrence and prognosis in esophageal cancer.

ABSTRACT

Objective To explore the antidepressant effect and mechanism of Chaihu Shugan San (CHSGS) composition, a compound traditional Chinese herb medicine and its components. Methods Rats were randomly divided into 6 groups: normal control group, model control group, a CHSGS group, a component I group, a component II group and a fluoxetine control group. The depression model was replicated by chronic unpredictable mild stress and single house for 28 d. Behavioral scores of the rats were detected by Open-field test and sucrose solution consumption test, and ERK1/2 mRNA expression in the hippocampus tissue was assayed by real-time fluorescent quantitative PCR. Results ERK1/2 mRNA expression was down-regulated in the depression model group compared with the normal control group (P<0.01). Compared with the model group, ERK1/2 mRNA expression in the CHSGS group and fluoxetine group was both up-regulated (P<0.05 or P<0.01); and only ERK1 mRNA expression in the component I group was up-regulated (P<0.05). No significant difference existed between the component II group and the model group (P>0.05). Conclusion Isolated-living condition and chronic mild unpredictable stress can down-regulate the expression of ERK1/2 mRNA in the hippocampus tissue. CHSGS may exert an antidepressant effect through increasing the expression of ERK1/2 mRNA in the hippocampus, component I may play an important role in its antidepression effect, while compatibility of the use of component II can enhance the antidepressant efficacy.

ABSTRACT

Objective To investigate the effects of Chaihu Shugan San(CHSGS) ,a compound traditional Chinese herbal medicine,on the behavior and plasma levels of corticosterone level and corticotrophin releasing hormone ( CRH) mRNA expression in the prefrontal cortex and the hypothalamus of mice with chronic mild unpredicted stress depression. Methods 40 adult male Kunming mice were randomly divided into 4 groups: the normal control group,the model control group,the fluoxetine group and CHSGS group. Except the normal control group, mice were exposed to an unpredicted sequence of mild stressor for continuous 21 days to induce depression. Since the 8th day, mice were intragastrically administered with equal volume agents respectively for 14 days (normal saline for the normal control group and the model group,fluoxetine(2. 6 mg · kg-1 ) for fluoxetine group and CHSGS (8.45 g · kg-1) for CHSGS group). Body weight and behaviors were investigated,the plasma levels of CORT in different groups were detected by radioimmunoassay, and the mRNA expression in the prefrontal cortex and the hypothalamus were semiquantified by the RT-PCR method. Results In comparision with the model group, body weight changes ( (13.22 ±3.57)g) and consumption of sucrose solution((1. 90 ±0. 74) ml) of CHSGS groups were significantly increased, and the duration of immobility during the forced swimming ((62.20±13.73)s) and tail suspension((58. 10 ± 11.71 )s) ,the levels of plasma CORT((12.61 ±4.63)ng/L) and the CRHmRNA expression in the prefrontal cortex(0.34 ±0.10) and the hypothalamus(0.70 ±0.09) were significantly decreased. Conclusion CHSGS can reverse the depressive behavior, reduced the levels of plasma CORT and down-regulated CRHmRNA expression in the prefrontal cortex and the hypothalamus of the depressive mouse caused chronic stress.

ABSTRACT

Objective: To investigate the effects of a Chinese herbal formula for calming liver and suppressing yang on the protein expressions of vascular tissues in spontaneously hypertensive rats (SHRs), and to explore the mechanism of efficacy. Methods: Twenty SHRs were randomly divided into model group and treatment group. Another 10 Wistar-Kyoto rats were selected as a normal control. SHRs in the treatment group were administered with the formula for calming liver and suppressing Yang for 4 weeks. During the course of treatment, blood pressure and heart rates were monitored every week and the ethology of rats, including irritability and rotation endurance was also evaluated. After treatment, thoracic aorta was obtained and its proteins were separated by 2-dimensional gel electrophoresis (2-DE). The differentially expressed proteins were identified by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) and database query. Results: The formula for calming liver and suppressing yang not only decreased the systolic blood pressure and heart rate, but also improved irritability degree and rotation endurance time of SHRs. This experiment had established the 2-DE graph of protein expressions of vascular tissues in SHRs. Compared with the normal group, the expressions of 15 proteins were down-regulated, and 12 proteins were up-regulated in vascular tissues of the model group. The formula for calming liver and suppressing yang treatment up-regulated expressions of 10 proteins in the 15 down-regulated proteins, and down-regulated 8 proteins in the 12 up-regulated proteins in vascular tissues of SHRs. After analysis, 16 obviously differentially expressed proteins were found, and 13 of them were identified. Conclusion: The formula for calming liver and suppressing yang can improve the ethology of SHRs. The mechanism is probably concerned with regulating the protein expressions of vascular tissues.

ABSTRACT

<p><b>OBJECTIVE</b>To observe the therapeutic effect of calming the liver and restraining the Yang formula in treating patients with mild or moderate degree of essential hypertension (syndrome of hyperactivity of liver-Yang), and to explore its mechanism in lowering blood pressure.</p><p><b>METHOD</b>The 348 patients with EH of stage I , II were randomly divided into two groups, the 174 patients in the treated group were treated with the calming the liver and restraining the Yang formula, and the 174 patients in the control group were treated with amlodipine. The treatment course for them all was 12 weeks. The related clincial symptoms score and quality of life score estimated before and after treatment at 4th week, 8th week and 12th week were observed. Before and after treatment, the ambulatory blood pressure (AMBP), heart rate, blood lipid, serum livels of high-sensitivity C-reactive protein (Hs-CRP), Angiotensin-II (Ang II) and calcitonin gene-related peptide (CGRP) were measured respectively in 40 patients of the treared group and 40 patients of the control group.</p><p><b>RESULT</b>After treatment, the treatment in the treated group showed an effect better than that in the control group in terms of nigh-time blood pressure reducing rate (P < 0.05). The reducing blood pressure variability and total effective rate in the treated group were no significant than that in the control group. In respect of reducing symptomatic scores on dizzy, soreness and weakness of the waist and knees, disturbed and dry and bitter of mouth, ameliorating quality of life score, decreasing the levels of heart rate, total cholesterol (TC), triglyceride (TG), high-density lipoprotein cholesterol(HDL-C) in the treated group were showing marked improvement as compared with that in the control group (P < 0.05 or P < 0.1). The improvement in the level of Ang II , Hs-CRP and CGRP before treatment in two groups were more significant than that after treatment (P < 0.05). However There were no difference in after treatment between the treated group and the control group.</p><p><b>CONCLUSION</b>The calming the liver and restraining the Yang formula shows favorable efficacy in lowering blood pressure on the patients with mild or moderate degree of essential hypertension. It can reduce the clincial symptoms, improve the quality of life, regulate blood lipid metabolism. Its mechanism may be related to the functional relieving inflammatory reaction and inhibition the activity of renin-angiotensin-aldosterone system (RAAS).</p>

ABSTRACT

Objective To evaluate the clinical effect of retrievable esophageal covered metal internal stent in treating patients with achalasia.Methods Under DSA guidance,peroral"Z-type"double horn covered metal internal stent implantation was performed in 16 patients with achalasia.Esophagography was carried out about 28 days after the procedure and the stent was retrieved.Results Of 16 cases,the stent fell off into the stomach two weeks after the operation in one.And the stent was successfully replaced after it was taken out.The placed stent was successfully retrieved in all cases 28 days after the treatment.No serious complications occurred.All the patients were followed up for 3 months to 3 years.During the follow-up period restenosis of the esophagus developed in two cases(at one and 1.5 years respectively),and the restenosis degree was relived after balloon dilation.Clinically,no esophageal symptoms,such as dysphagia,occurred in all patients.Conclusion As a simple and safe technique,the retrievable esophageal covered metal internal stent implantation is very effective with fewer complications for the treatment of achalasia.Moreover,the technique carries lower restenosis occurrence.

ABSTRACT

Objective To explore the effects of Tongfu Huayu Daotan Decoction (通化阏导痰汤)on the serum concentration of S-100β protein, neuron-specific enolase(NSE) and Prognosis in patients with severe craniocerebral injury.Methods Sixty patients with severe craniocerebral injury were randomly divided into a a'eatment group and a control group.The treated group was treated with Tongfu Huayu Daotan Decoction plus conventional treatments including dehydration,antibiotics, organ functional support, nerve nutrition, prevention of complication, etc.; the control group was treated with conventional treatments alone. The concentration of serum S-100β protein and neuron-specific enolase(NSE) in plasma at admission and at24, 36, 72huors, and 5, 7 days after treatment were determined respectively; the Glasgow outcome scale (GOS)and neurological deficits scoring at 2weeks and 4weeks after hospitalization were compared to observe the efficacy of the patients. Results The concentration of serum S-100β protein and neuron-specific enolase (NSE) in plasma at previous treatment and at 24, 36 hours after the treatment had no statistical difference in the two groups(P>0.05 ), The concentration of serum S-100β protein and neuron-specific enolase (NSE) in plasma at 72huors, 5 and 7 days after the treatment in the Tongfu Huayu Daotan Decoction group were lower than those in the control group, the differences being significant (P<0.01). The Glasgow outcome scale (GOS) and neurological deficits scoring at 2weeks and 4weeks after the treatment in the Tongfu Huayu Daotan Decoction group were significantly nigher than those in the control group, the differences being significant (P<0.01) .Conclusion Tongfu Huayu Daotan Decoction can alleviate the plasma concentrations of S-100β protein and neuron-specific enolase (NSE) in patients with severe craniocerebral injury and markedly improve the clinical therapeutic effects. Combined Tongfu Huayu Daotan Decoction and western medicine can significantly reduce mortality and improve the Glasgow outcome scale (GOS), neurological deficits scoring and therapeutic effect.

Methods Thirty-two bump brain injured patients with cerebral hernia were treated with inside and outside decompression of standard large trauma craniectomy compared with 24 cases of routine temporoparietal craniectomy between Oct 2001 and Aug 2004.Results In standard large trauma craniectomy group,21 cases got good recovery(65.6%) and 5 were dead(15.6%).In routine temporoparietal craniectomy group,9 cases got good recovery(37.5%) and 10 were dead(41.7%).There was significant difference between the two groups in good recovery and mortality(P

ABSTRACT

Objective To estimate the efficacy of super-early minimal invasive evacuation of hematoma via keyhole for treating hypertensive basal ganglionic hemorrhage. Methods 36 patients with hypertensive basal ganglionic hemorrhage(9 cases in grade Ⅱ, 17 in grade Ⅲ and 10 in grade Ⅳ) were treated by minimal invasive evacuation of hematoma via keyhole at super-early stage, and their efficacy was statistically compared with 35 patients with hypertensive cerebral hemorrhage underwent conservative therapy or lateral ventriculopuncture. Results In the 36 patients underwent minimal invasive evacuation of hematoma via keyhole, 22 cases had a good prognosis(61.1%), 9 had a poor prognosis(25.0%), and 5 died(13.9%). Among the 35 patients underwent conservative therapy or lateral ventriculopuncture, 5 cases had a good prognosis(14.3%), 22 had a poor prognosis(62.9%), and 8 died(22.9%). ABSTRACT

ve To study the way of oxidative stress induced by soot rat pulmonary cells type Ⅱ ?Methods Before exposure to soot extracts, the rat pulmonary cells were pretreated with anti-oxidative agent, N-acetylcysteine (NAC), vitamin C, vitamin E and mannitol for 30 minutes, then the ceils were exposed to the soot extracts for 24 hours. MTT assay was applied to detect the cytotoxicity. Results It showed that NAC, vitamin C and vitamin E could reduce the cytotoxicity of rat pulmonary cell caused by soot, but no same change was observed in that of man-nitol. Conclusion It was suggested that the DNA oxidative stress of soot may be caused by multi-ways, such as in-duction of oxygen free radical and lipid peroxide etc.
